Abstract

Well-directed treatment of tumors with therapeutic genes is the aim in the development of modern cancer therapies. While successful trials in viral gene therapies are always associated with the possibility of severe drawbacks, it is necessary to work on nonviral alternatives. Based on our experiments, specific enhancement of liposomal transfection is possible in cells from gynecological malignancies when they express high amounts of steroid receptors. By introducing suicide genes into these specific kinds of cells, targeted and efficient chemotherapy could be achieved in gynecological malignancies that is followed by less side effects. With the addition of steroids to the liposomal transfection, we obtained a cell-specific enhanced expression of the applied reporter gene luciferase. When steroid receptors migrate into the nucleus after their activation by steroids they might co-transport the exogenous DNA.We followed this hypothesis to develop a targeted enhanced transfection in gynecological malignancies expressing high levels of steroid receptors.
